Majority of the cereals are highly recalcitrant to Agrobacterium-mediated transformation, and so direct transfor-mation methods have been developed to transform such plants. Which of the following direct transformation methods is applicable to intact plant tissues?
-
- Calcium chloride and PEG-mediated transformation
- Liposome-mediated transformation
- Electroporation
- Transformation using microprojectiles
Correct Option: B
Lipofection (or liposome transfection) is a technique used to inject genetic material into a cell by means of liposomes, which are vesicles that can easily merge with the cell membrane since they are both made of a phospholipid bilayer. Lipofection generally uses a positively charged (cationic) lipid to form an aggregate with the negatively charged (anionic) genetic material and hence is the best mechanism to transform intact plant tissues.
